Identification of cytokinins associated with mitosis in synchronously cultured tobacco cells

The amount of endogenous cytokinins in the butanol-soluble fraction increased about 5 times during the mitosis of synchronously dividing cultured tobacco cells (Nicotiana tabacum cv. Xanthi). The free cytokinins were identified as trans-zeatin and zeatin ribonucleoside by paper and thin layer chromatography and by high-performance liquid chromatography combined with mass spectrometry and bioassay. The water-soluble fraction contained zeatin ribonucleotide, isopentenyladenosine monophosphate, phosphoglucosylzeatin and phosphoglucosylzeatin ribonucleoside. Changes in the cytokinins in the water-soluble and transter RNA fractions during the cell cycle did not support the hypothesis that they might be the source of the butanol-soluble cytokinins which increased during mitosis. These seem to be synthesized de novo during mitosis.
