Extraction of rapidly labeled RNA from broad bean shoots and tobacco leaves was investigated by using several extraction media ordinarily combined with phenol. Three DNA-like RNA classes and an rRNA-type class have been identified in the rRNA regions of methylated albumin-coated kieselguhr (MAK) column chromatographic profiles from both plants. One DNA-like RNA class of high molecular weight is almost inextractable without the addition of an alkaline agent or a detergent, while the other two size classes can be extracted effectively with or without additives, suggesting the possibility of their possessing different forms of migration from nucleus to cytoplasm. In cytoplasm, two rather distinct ribonucleoprotein complexes of about 35 S and 62 S were detected, which contained two size classes of DNA-like RNA.
