Enzymes of the C_4, C_3 pathway and photorespiration have been analyzed for P. hians and P. milioides, which have chlorenchymatous bundle sheath cells in the leaves. On whole leaf extracts the levels of PEP carboxylase are relatively low compared to C_4 species, RuDP carboxylase is typical of C_3 species, and enzymes of photorespiratory metabolism appear somewhat intermediate between C_3 and C_4. Substantial levels of PEP carboxylase, RuDP carboxylase, and photorespiratory enzymes were found in both mesophyll and bundle sheath cells. Low levels of C_4-acid decarboxylating enzymes may limit the capacity for C_4 photosynthesis in P. hians and P. milioides. The results on enzyme activity and distribution between mesophyll and bundle sheath cells are consistent with CO_2 fixation via C_3 pathway in these two species.
