(1) To analyze the photoelectron flow related to photosystem II, particle II preparation, i.e., the chloroplast fragment having only photosystem II activity, proved to be far better than the generally used chloroplast preparations having activities of both PS-I and PS-II. (2) By simultaneous measurements of the activities of O_2 evolution and DPIP- and ferricyanide photoreduction using variously-treated particle II preparations, it was found that a noticeable activity of ferricyanide photoreduction was still observed, though the former two activities were completely lost in the course of treatments such as Tris-treatment, pre-illumination and aging. (3) Besides this, differences were found between ferricyanide- and DPIP-photoreduction in respect to susceptibility to CCCP, availability of artificial electron donor, and the effect of chloride addition. However, both photoreductions were equally inactivated by heat-treatment and addition of DCMU. (4) To explain the observed distinctions between DPIP and ferricyanide in their mode of action as electron acceptor for PS-II, a scheme suggesting the involvement of two light reactions in PS-II is proposed and the electron flow near PS-II is discussed.
