1) Purification of the photoactive particle corresponding to photosystem II (particle II) was achieved using a combination of procedures including digitonin- and Triton X-100-treatments, sonication, and differential- and density-gradient centrifugations. 2) The "purified" particle II preparation showed only oxygen evolution activity and showed no NADP^+ photoreduction activity even when an electron donor couple was added. 3) The chemical compositions of this particle and of the particle corresponding to photosystem I (particle I) were compared with respect to the composition and contents of their chlorophylls, carotenoids, cytochromes, plastoquinones, protein and lipid. Marked differences were found. 4) Using the two photoactive particles I and II lacking the respective counterpart activity, a partial success was attained in reconstituting a system, in which photoinduced electron flow from water to NADP^+ was observed, provided that suitable intermediate electron carriers, e, g, plastoquinone and plastocyanin, were added. 5) The nature of the two photoactive particles obtained and their relationship to particles so far reported are discussed.
