The thread-like apparatus and its movement are observed clearly with a phase contrast microscope, as shown in the photomicrographs. When caffeine solution is used as a medium, the thread-like apparatus and the tannin vacuole have a precipitation in them. There is some degree of caffeine reaction in these threads. A part of the thread-like apparatus assumes a bead-like form or a small mass, and the rest of the thread becomes long and slender. Tiny granules are observed near thread-like apparatuses with a phase contrast microscope, but these granules cannot be revealed with vital staining. The vermiculation of the thread-like apparatus is slower near the nucleus than in the vicinity of chloroplasts. Generally speaking, the movement of the thread within the living cell can be said to be marked.
