In the present investigation, the changes in the tissue-cells of Mimosa padica L. under diurnal and noturnal conditions were studied from morphological and cytophysiological points of view. The results obtained may be summarized thus: 1) Tannin substances or tannin vacuoles in the plant tissue can be fixed with Kaiser's fluid, and a 0.5 per cent alcoholic solution of toluidin blue has been discovered to be a specific agent for staining the tannin. 2) Kaiser's fluid-toluidin blue method has been found to tinge the motor tissue differently before and after the stimulus is received. 3) Tannin substances infiltrate the cytoplasm after the stimulus is received under diurnal donditions. Moreover, in this condition a thin cytoplasm is frequently observed. 4) Under nocturnal conditions, after the stimulus is received the mixing of tannin and cytoplasm is observed in some parts of the motor tissue. In this case, then, dilute tannin substances issue from the cell. 5) In fresh material, the thread-like apparatuses under diurnal conditions are thicker than those in the noctnrnal period. The threads have a tendency to be fixed by the Champy-Toriyama method as a bead-like feature under diurnal conditions. On the contrary, in the nocturnal period, the threads appear to be thin and slender. 6) By the Champy-Toriyama method, the tannin vacuoles in the tissue-cells under diurnal conditions have been observed to be more inflated than those under nocturnal conditions. Such a difference has also been observed in fresh material. 7) Fixed with neutral formalin, the carotenoid-body is markedly stained a brilliant cresyl blue. Carotenoid-bodies are uniformly distributed throughout the tissue-cells of the main pulvinus, the petiole and the mesophyll of the leaf. 8) By the author's method, carotenoid-bodies in the diurnal period have been observed to be more inflated than those in the nocturnal period. 9) Judging from the data obtained by these observations, it may be concluded that the carotenoid-body is a cellular element and that itis by no means a mere artifact.
