Effect of reserpine pretreatment on the lowering of skin temperature, resulting from contraction of peripheral blood vessels by norepinephrine and ethylphenylephrine, was examined by the measurement of ear temperature in uninjured rabbits. Rabbits were injected subcutaneously with 5 mg/kg of reserpine 24 hr before the experiment. Lowering of the ear temperature in rabbits given either of the two drugs was stronger and lasted longer than that in nontreated rabbits. The dose-reaction relation (area below the time-lowering degree curve) shifted towards the left by the two drugs in the treated animals, and the lowering action was 2-fold stronger in norepinephrine and ca. 5-fold stronger in ethylphenylephrine than in nontreated animals. These results indicate that pretreatment with reserpine fortifies the lowering action when sympathomimetic amines (vasoconstrictors) are evaluated with lowering of ear temperature as the index.
