The aim of this study is to elucidate the metabolism of RNA and DNA during the interphase of Paramecium caudatum. The stock culture was transferred into the physiologically balanced solution containing ^<32>P. After incubation at 25℃ for 1.5 hours, organisms were fixed, a series of histological sections being prepared. The preparations were then treated with RN-ase or DN-ase to remove RNA or DNA respectively. The microautoradiograms were made by means of stripping method and then the numbers of the tracks were counted. Meanwhile the nucleic acid contents in macro- and micronuclei as well as the cytoplasm were determined after the differential centrifugation. The result of chemical analysis revealed that little RNA was detected in micronuclei whereas both nuclei contained an appreciable amount of DNA. However DNA was found metabolically inactive throughout the interphase. On the contrary, ^<32>P was incorporated into both RNA found in macronuclei and cytoplasm actively, especially during intermediary stage from daughter cells to the prefission stage.
