1. The distribution of acidic and alkaline phosphatases in the intestinal cells of human and pig ascarids were investigated. The human ascarids were discharged by santonin and the pig ascarids were surgically taken out from the small intestine of the host. 2. The specimens were fixed according to Moog ('44), with chilled acetone. Acidic and alkaline phosphatases were studied by the methods of Moog ('44) and Gomori ('39, '41) respectively, using sodium glycerophosphate as a substrate. 3. Both phosphatases are present only in the mid-gut which constitutes the major part of alimentary canal. There is no difference in this mode of distribution of phosphatases between human and pig ascaids. 4. The reaction of acidic phosphatase is particularly intense in the striated cuticular border which faces to the alimentary tract. The acidic phosphatase is also present in the subjacent hyaloplasm, and in the nucleus and the supra-nuclear region as well. In the supra-nuclear region the reaction of acidic phosphatase seems to be connected with the small spherical granules which are lacking in other parts of the cells. As to the nucleus the stain is detected on the surface and the granules therein. When fully given reaction, a net-work existing in the basal region of the cells becomes to present a positive reaction. But the basal membrane of the mid-gut is always negative. 5. Comparing with the acidic phosphatase, the alkaline phosphatase is quantitatively small, and requires more than 24 hours incubation for its detection. The reaction is always found to be connected with the spherical bodies in the cells. Sometimes, however, the same reaction is found in the striated cuticular border, subjacent hyaloplasm and nucleus, but the reaction of these parts is quite irregular, the reason is unknown. 6. The distribution of two kinds of phosphatase and the degree of their reactions to the dyes change in close relation to the conditions of the intestinal cells. Further details will be made the subject of the future report.
