1. The present experiments were carried out with the purpose of investigating the factors governing laterality, especially those underlying the determination of the antero-posterior axis (ap-axis), of the homoiogenetically induced limbs. 2. The laterality of the induced limbs obtained in experiments, where a limb bud at stages 1/1-1.5/1 of Bufo vulgaris was grafted to Hynobius tokyoensis at the stages 38 and 41, was determined principally by noting the position of the limb relative to the transplant and the normal host limb. The relative position of the induced limb to the transplant and the orientation of the transplant were closely related. 3. In the aa. dd-transplantations the induced limbs occurred at a position immediately posterior to the transplant, i.e., between the transplant and the normal hind limb and they were disharmonic (polarity, ap. dd). On the other hand, in the ap. dd-trnasplantations the induced limbs were formed just anterior to the transplant, when seen from the host hind limb, and they were harmonic (polarity, aa. dd). Accordingly, it was found that the ap-axis of the induced limb was determined by the relative position of it to the transplant and the normal host limb and also by the orientation of the transplant. 4. In the present experiments the effect of altering the axial relationship between the limbs on the laterality of the induced limb was studied by the following operation. After the transplantation of a limb bud of Bufo, (1) in one group the transplant was removed from 3 to 20 days after the operation, and (2) in anoter group a second transplant was placed between the first transplant and the normal hind limb on various days, ranging from day of the first transplantation up to 7 days after. 5.When the first transplant was removed at an early time, the induced limb either was not formed or did not develop well and it was found that the transplant must remain for more than 7-9 days in the host body in order to yield the induced limb. 6. The laterality of the limb induced by the first transplant was disharmonic in the aa. dd-transplantation and harmonic in the ap. dd-transplantation, regardless of any change in the relative position of the transplant and the host hind limb following the removal or addition of the transplant. Accordingly, there is a strong indication that the ap-axis of the xenoplastically induced limbs is polarized as soon as the limbs determined to be formed and that it could not be changed by the second operation.
