With a hope of finding a tissue culture method available for lower vertebrates, monolayer tissue culture techniques and different culture media formulae were followed with various internal organs of amphibians. It was found that the monolayer culture technique used for mammalian tissues was also useful in the culture of tissues of two anurans, Hyla arborea japonica and Rana temporaria. For tissues of the newt, Triturus pyrrhogaster, the plasma clotting method was useful in the growth of cells. Cultures were successfully made in tissues from young frogs as well as from adult frog hearts. Cells of the tree frog, Hyla, showed a generally better growth than those of the frog. In newts, cell growth was essentially dependent on the existence of the chicken extract in the medium. Chromosomes were studied in kidney cells of Hyla grown in culture. It was found that in males the diploid number (2n) was 24. Two points of special interest: the male cell showed a pair of unequal size which is expected to form an X-Y complex; and the 6th pair consisted of members showing a secondary construction in each short arm.
