肝マイクロボディの比較形態学的研究 : II. 海産軟体動物および甲殻類肝膵臓のマイクロボディと酵素活性

Previously, the existence of microbodies in hepatopancreas of the marine mollusca and crustacea was demonstrated by the present author. This work dealt with the relationship between these microbodies and the enzyme activities of hepatopancreatic homogenate. Catalase, peroxidase, and urate oxidase were assayed. Catalase activity was demonstrated spectrophotometrically in mollusca but not in crustacea. Peroxidase activity was observed spectrophotometrically in both mollusca and crustacea. Urate oxidase activity was not confirmed in hepatopancreas of borh mollusca and crustacea, but detected by the same method in liver of fish which was collected at the same time. From these results, it seems that the hepatopancreatic microbodies of mollusca, which were stained with the diaminobenzidine (DAB) reaction, contain both catalase and peroxidase, and that the crustacean hepatopancreatic microbodies possess only peroxidase since catalase activity is absent in the homogenate. The previous observations suggested the possibility that the microbodies containing nucleoid might exhibit urate oxidase activity according to a hypothesis of Shnitka (1965). However, it appears that the hypothesis cannot be applied to the microbodies of the animals used in this study. Electron microscopic observations carried out in the same way as the previous study showed again that the microbodies were contained in the hepatopancreas of all the species (mollusca: 2, crustacea: 6) employed in the present study. Of these animals, the microbodies of a hermit crab, Panulirus japonicus showed negative reaction of DAB. Nucleoid was seen in a bivalve, Spondylus barbatus and a crab, Xantho (Leptodius) exaratus. In the former the nucleoid exhibited a lattice pattern and in the latter it was an electron dense mass. From the results obtained in this study, it may be suggested that the molluscan hepatopancreatic microbodies are similar to mammalian hepatic microbodies, but crustacean microbodies are different from those of mammalia in their enzyme composition.
