Cells with double nucleus have been frequently observed in various embryonic tissues and have been assumed to be caused by amitosis. In our observation on isolated embryonic cells of Japanese Medaka, Oryzias latipes, it was not found to be the case. Embryonic cells can be easily dissociated in M/15 Sorensen's buffer solution in which cells continue to divide as in normal saline solution for several hours. In the normal course of mitosis hyaline pseudopodia (lobopodia) appear on both surfaces of the dividing cell which move toward the cleavage furrow and they push against each other to accomplish the final stage of the cytokinesis. In some cases only a single lobopodium is formed on certain place of the cell surface and moves around the cell surfaces. Then the cell returns to its original rounded shape with two nuclei, hence the double nucleus is formed by "abortive mitosis" and not by amitosis. After a while this cell again divides into two mono-nucleated cells, and, as Sirakami (1958) has suggested, the double nucleus is by no means pathological phenomenon. In this report various types of the double nucleus formation were described.
