Fushion has been induced during mechanical isolation of Oryzias embryonic cells in normal saline solution (0.75% NaCl, 0.02% KCl, 0.02% CaCl_2, 0.02% NaHCO_3) by quickly bringing about physical contact between cells. The more rapidly the isolated cells were brought into contact, the more the cells performed fusion. About half of the paired cells fused by rapid production of cell-to-cell contact within about 20 sec of cell dissociation. If individual cells remained isolated for about 90 sec, cell fusion could no longer be induced. A frequent fusion could be induced in Ca^<++>-free saline solution, in Ca^<++>-free+0.02% MgCl_2 saline solution and in Sorensen's phosphate mixture (pH 7.7). In Ca^<++>-free+0.1mM EDTA saline solution, however, the fusion ratio decreased. Various pH (6.0, 7.4, 9.0) of the saline solution did not affect the fusion ratio. Frequent fusion could also be induced in saline solution of lower or higher osmotic pressure.
