1. Observations on the relation between the formation of cortical alveoli and the yolk-nucleus were carried out in the ovarian and mature eggs of the fish, Plecoglossus altivelis. 2. The cortical alveoli originate in the yolk nucleus or its fragments. During the growth period the yolk nucleus forms several fragments. The basophilic fine granules appear first around the yolk nucleus and its fragments, and then disperse gradually in the whole cytoplasm. Increasing somewhat in size, the scattered granules aggregate and fuse to form many small vacuoles, which grow into the cortical alveoli later. During this process the granules or their derivatives decrease in stainability with basic dyes, while increasing in the reaction with PAS-reagent. 3. The fragmentation of the yolk nucleus occurs several times in course from its first appearance in the cytoplasm up to the migrating period of the germinal vesicle towards the animal region of the oocyte. The basophilic substance in this oocytes seems to be identical with the "pallial substance or pallial layer" described previously, judging from its behavior, stainability and shape. 4. The minute yolk granules having a extremely weak stainability with the iron-haematoxylin appear first in groups around the germinal vesicle. After a slight increase in their stainability and volume they accumulate to form a number of small ring-like bodies which grow into yolk globules with a gradual increase in their volume.
