Experiments were carried out to get information about the source of calcium involved in initiating caffeine contracture of a molluscan catch muscle (anterior byssal retractor muscle of Mytilus edulis LINNE). Throughout the experiments, bundles of fibres (0.3-0.7mm in diameter) obtained from the muscle were used. In artificial sea water containing a sufficient amount of caffeine the muscle bundle contracted strongly without any appreciable change in the transmembrane potential. The caffeine contracture was potentiated with an increase in the external calcium concentration. This potentiating effect reached its maximum within 5 to 15 minutes after the increase in the concentration, and remained at that level. The potentiating effect of the solution containing high calcium lasted for a long time (more than an hour) after the concentration had returned to normal, and was eliminated completely by the application of caffeine. In artificial sea water to which no calcium was added, the first caffeine contracture was still brought about 10 hours or more after immersion of the muscle bundle in this solution, but the second contracture was greatly reduced or completely abolished only one and a half hours after the immersion. In NaCl (556mM)+EGTA(1-3mM) solution which is isotonic with artificial sea water, the first caffeine contracture could be obtained until about 6 hours after immersion of the muscle bundle in this solution, but the second contracture was not brought about only one and a half hours after the immersion. These results mentioned above suggest that there are sequestered-calcium binding sites in the muscle cell, and caffeine may act to release this bound calcium into the cytoplasm and initiate a contracture.
