Effects of some divalent cations (Sr^<++>, Mn^<++>, Co^<++>, Ni^<++>), serotonin (5-hydroxytryptamine), and procaine on the caffeine (10mM) contracture of a molluscan catch muscle (anterior byssal retractor muscle of Mytilus edulis LINNE) were studied. Throughout the experiments, bundles of fibres (0.3-0.7mm in diamter) obtained from the muscle were used. The caffeine contracture was potentiated by an addition of 30mM of heavy metal ion (Mn^<++>, Co^<++>, or Ni^<++>) to external solution. The potentiating effects of these metal ions reached their maximum about 10 minutes after the addition, and then decreased. Thirty minutes after the addition, the peak tension of each contracture was found to be a little esmaller than those obtained in normal solution. Sr^<++> (30mM) also potentiated the contracture. The potentiating effect reached its maximum about 15 minutes after the addition of the ion, and then decreased gradually though the decrease was markedly slow as compared with those in the cases of heavy metal ions. The potentiating effect of Sr^<++> lasted for a long period of time (more than 50 minutes) after the ion had been withdrawn. On the contrary, the potentiating effects of heavy metal ions were completely eliminated within 20 minutes after the withdrawal of the metal ions. Following abolition of the caffeine contracture of the muscle bundle in the solution to which no Ca^<++> was added, none of the heavy metal ions (11mM) could restore the contracture, but Sr^<++>(11mM) could restore it. Both serotonin (10^<-6>M) and procaine (1mM) reduced the peak tension of the contracture to about one third of the normal when each drug was applied to the muscle bundle with caffeine. When each drug was applied to the bundle after the contracture had developed into its maximum, however, procaine had no significant effect on the relaxation rate, while serotonin greatly increased it. The modes of action of these divalent cations of drugs were discussed.
