The metabolism of ecdysone in the pupal development of the fleshfly, Sarcophaga peregrina, was studied using ^3H-ecdysone Ecdysone was rapidly metabolized and converted mostly into three groups of metabolites, OA, OB and unconjugated ecdysteroids similar to those obtained injection of ^3H-20-hydroxyecdysone. Though components of ecdysteroids contained in the conjugated were similar to unconjugated metabolites, there was a difference in take rates of the components of metabolized ecdysteroids. The main ecdysteroids metabolites were supposed to be 25-deoxy, 20, 26-dihydroxy-ecdysone, 26-hydroxyecdysone and 20, 26-dihydroxyecdysone judging from the development of TLC and from the elution pattern of standard samples of ecdysteroids by HPLC. Radioactive 20-hydroxyecdysone was scarceiy at any time from 1 to 72 hr after injection of labeled ecdysone. We found that in the pharate pupal stage, at a time when the ecdysteroids titer was rapidly decreasing, the injected ecdysone was immediately converted to inactivated metabolites instead of producing 20-hydroxyecdsone.
