Purpose: The 1C8 antibody was prepared as a monoclonal antibody against nerve growth cone compartment from the rat brain. On histochemical study using the 1C8 antibody in the rat brain and gastrointestinal tract, positive staining was observed in ileum, duodenum, and brain. Immunoelectron microscopic analysis of ileum goblet cell showed that the antigen is on Golgi apparatus and mucous granule membrane. These results suggest the 1C8 antigen play a role both brain and intestine. In this work, biochemical study was performed to characterize the 1C8 antigen. Methods: Triton X-100 extract from brain membrane fraction was done by SDS-PAGE and blotting to membrane. The membrane proteins treated with and without N-glycosidase F or sialidase were immunostained. In intestine, two membrane fractions were prepared Results: In brain, the 1C8 antigen was 25kDa protein. The immunoreactivity was disappeared by the sialidase treatment and not changed with N-glycosidase F treatment. In intestine, immunoreactive protein was not detected. Discussion: In brain the epitope may exist on the O-type sugar chain of the glycoproteins. In intestine, the 1C8 antigen may be the glycolipid. In the general pathway of carbohydrate biosynthesis the sialic acid residure transferred on Golgi apparatus. It is correspondingly considered the antigen processing the pathway.
