Mice were injected with lymphocult T, an activator of T-cells, into the gingiva. T-cells were isolated from submandibular lymph nodes and spleen separately at 7th, 10th and 14th days after the injection. T-cells isolated were tested for both cytotoxity and Fc receptor positiveness and the proportion of each subclass was calculated. Monoclonal antibodies of Lyt 1, Lyt 2 and Lyt 3 were used in the cell selection of the former test, and IgM and IgG were used in that of the latter test. Results obtained were as follows : 1. In submandibular lymph nodes, the percentage of Lyt 1 antigen positive cells decreased from 23.8 to 16.1 during the experimantal period. In the cases of Lyt 2 and Lyt 3 antigens both percentages decreased from 22.6 to 14.9 and from 13.9 to 11.5, respectively. 2. In spleen, the percentage of Lyt 1 antigen positive cells decreased from 17.3 to 16.2. In the cases of Lyt 2 and Lyt 3 antigens both percentages decreased from 22.6 to 14.9 and from 13.8 to 6.9, respectively. 3. In submandibular lymph nodes the percentage of helper T-cell was calculated from Lyt 1, 2 and 3 positiveness as the range 17.3 to 16.3 and that of suppressor was 43.7 to 36.2. 4. In spleen the percentage of helper T-cell was caculated from Lyt 1, 2 and 3 positiveness as the range 17.3 to 16.3, respectively. Suppressor T-cell was dominant in number as compared with helper one in spleen. 5. The number of helper T-cell was recognized in the same or more by the calculation of positiveness of FC receptor as compared with that of suppressor T-cell in submandibular lymph nodes and spleen. 6. The number of cell matured to the specific subclassed one were more frequently observed in submandibular lymph nodes than that in spleen. Thse result showed that the cytotoxic test was more sensitive than the test of Fc receptor positiveness in the study of the classification of subclass of T-cell.
