Malignant tumors were induced in the cheek pouch of hamsters by daily application by stabbing of 0.5% solution of 9, 10-dimethyle-1, 2-benzanthracene (DMBA) in acetone, Gross and histologic investigations were made of the process of carcinogenesis and the induced tumors. Allografting of the induced tumors was experimented and lines of successively transplanted tumors were prepared. The results were as follows : 1. The process of carcinogenesis in gross observation consisted of redness-swelling stage, ulceration stage, healing stage of ulcer scar, formation stage of tumor, and proliferation stage of tumor. 2. Gross types of tumors were mainly papillary type (78.8%) and broad elevated type (21.2%). The first stage showed papillary type and broad elevated type was observed after 15 weeks. 3. Histopathologic process of carcinogenesis consisted of starting stage of experiment, ulceration stage, stage for proliferation of granulation tissue and regeneration of epithelium, formation stage of fibroid polyps, papilloma formation stage, stage tor carcinogenesis of papillary type, stage for carcinoma invasion with surface necrosis, and carcinoma of broad elevated type. However, generation process of malignant fibrous histiocytoma (MFH) and fibrosarcoma could not be observed. 4. Sixty-six cases of induced tumors consisted of 49 cases (74.2%) of squamous cell carcinoma, 11 cases (16.7%) of composit tumor (carcinosarcoma) of squamous cell carcinoma and MFH, 4 cases (6.1%) of isolated MFH, and 2 cases (3.0%) of coexistence of squamous cell carcinoma and fibrosarcoma. MFH numbered 15 when cases isolated or in composition with squamous cell carcinoma were combined. 5. Malignancy, in accordance with Broders' classification, of squamous cell carcinoma was grade 1 in 9 cases (18.4%), grade 2 in 29 cases (59.2%), grade 3 in 9 cases (18.4%), and grade 4 in 2 cases (4.1%). Accordig to Willen's classification, the most malignant case had a total of 23 points and the least malignant case a total of 7 points. 6. Metastasis to the submandibular lymph node was seen in 5 cases and all cases were squamous cell carcinoma. No cases of metastasis to the lymph node of MFH or fibrosarcoma were seen. 7. Histopathologic form of all MFH cases (15 cases) was predominantly pleomorphic subtype of common type. 8. Fibrosarcoma in 2 cases was observed in the lower connective tissue of squamous cell carcinoma. 9. Allografting was tried out with use of tumor tissue from 10 of the induced tumor cases three types of tumors were successfully transplanted to 4th generation : i) MFH-47 line : the primary tumor was isolated MFH and its histologic form was well maintained after transplantation. ii) MFH-74 line : the primary tumor was a composite tumor (carcinosarcoma) of squamous cell carcinoma and MFH. After transplantation to 1st generation, only MFH was successively transplanted. With successive transplantations, storiform pattern became indistinct. iii) SCC-92 line : the primary tumor was moderately differentiated squamous cell carcinoma of papillary type. Histological changes after successive transplantations were not especially pronounced.
