Osteoclasts, which were isolated from femurs of one day-old rabbit, were cultured on human dentine-slice for one day. These cells and slice surface were observed by scanning electoron microscope. The results were as follow : 1. Osteoclasts and micropits were 11±2 and 20±6, respectively, per one slice. 2. Most of osteoclasts were round, and their diameters were in a range of 20-100μm. 3. Some osteoclasts had many pseudopodia at the edge of the cell, and others had flat lamellipodia at the edge. There were many microvilli, about 0.3μm in thickness and 0.2-1.0μm in length, on the central surface of cytoplasm. These microvilli decreased in number in proportion to the distance from the center of that surface. 4. Micropits were found on the surface of tooth sections where there were partially destroyed and exposed osteoclasts surrounded by lamellipodia. The exposed micropits appeared about 5μm inside the cell edge. 5. The micropits were scattered on the surface of the slice ; some appeared alone and others were grouped together. Each episode of the micropits was in a renge of 10-50μm in diameter. 6. Most of the micropits had fibrillar structure inside. The thickness of the fiber was about 2μm. There were several types of the excavations differing in depth : very shallow one with dense fibers, moderate one with sparse fibers, and deep one without any fibers. From these results the author discuss the morphology of osteoclasts and the mechanism of osteoclastic bone resorption.
