Bleomycin (BLM) and Mitomycin C (MMC) which are most frequently used in chemotherapy for oral cancer were selected, and each was combined with Vitamin C (VC) in administration. Effect of combined use of BLM and MMC with Vitamin C was histopathologically investigated in terms of side effect. The results were as follows : 1. In the terminal portion of the parotid gland, as compared with the administration cases of BLM (2.5 mg/kg/day) alone and of MMC (0.3 mg/kg/day) alone, the combined administration cases of BLM-VC and of MMC-VC both showed atrophy and disappearance of cytoplasm, pyknosis, deformation, and furthermore destruction of nuclei in acinous cells markedly. Along with these regressive changes, PAS positive substance decreased markedly. 2. In the epitherial cells of intercalated duct and of striated duct, as compared with the administration cases of BLM alone and of MMC alone, BLM-VC combined cases and MMC-VC combined cases showed atrophy of cytoplasm, pyknosis and deformation of nuclei. These regressive changes however were slighter than those in the terminal acinous cells. 3. In the secretory duct, morphological changes were hardly seen throughout the experiment period. 4. The foregoing regressive changes were seen in the peripheral part of the gland and gradually extended into the central part and were intensified in degree with increase in the number of administration. 5. On the basis of the foregoing experimental results, BLM and MMC both affected not only morphological regressive changes but also functional aspects such as synthesis, accumulation and excretion of secretory substance. Furthermore, the combined use with VC was thought to aid and promote these regressive changes and decline in function.
