実験的歯周炎の細胞間相互作用によるインターロイキン 2 レセプター (IL-2R) 発現に関する研究

It is known that activated T cells play an important role in cellular immune response in lymphocyte-rich early periodontal lesion. Activated T cells produce a lymphokine, interleukin-2 (IL-2), which binds to IL-2 receptors (IL-2R) and exerts a multiplicity of biological activities modulating many immune responses. The purpose of this study is investigating the role of IL-2R in intercellular activating reaction in periodontal lesion in vivo and in vitro. In in vivo experiment, cell activators, lipopolysaccharide (LPS) and interleukin-1α (IL-1α) were injected into gingiva of rats. Cell infiltration of gingiva appeared in all specimens injected with the activators. IL-2R were observed at the same site of macrophages (Mφ) antigen positive cells in all thin sections at an area of cell infiltration. When T cells were attracted to gingiva by ConA injection and then were activated by LPS injection, IL-2R expression, Mφ accumulation and cell infiltration were intense compared with that in the rats LPS or IL-1α injection alone. The result suggested that Mφ were activated and expressed IL-2R through the interaction with T cells in cell infiltrating area. To examine the effect of T cells on Mφ IL-2R expression during cell activation in vitro, LPS sensitized T cells and Mφ were incubated in the presence of activators, LPS and IL-1α. IL-2R expression on Mφ depended on sensitized T cells density and incubating period in the culture with LPS. IL-1α also makes express IL-2R on Mφ in the culture using normal T cells and Mφ. IL-2R expression is two fold higher when Mφ incubated with sensitized T cells, instead of normal T cells. This result suggested that sensitized T cells secreted lymphokines which may act on Mφ to amplify the immune response in a sequential fasion by establishing a positive feedback circuit. Above results suggested that IL-2R plays an important role in an immune response involving the interaction between Mφ and T cells in early periodontal lesion.
