Experimental pulpitis by inoculating lipopolysaccharide (LPS) into root pulp was studied in 2 groups of rats with one group immunized previously with LPS. The effect of immunization on pulpitis was discussed. The rats of both groups were sacrificed in the 1st, the 2nd, the 3rd, the 4th, and the 5th week after the inoculation. Sera, teeth and periodontal tissues were prepared from the animals. The concentration of immunoglobulins (Ig's) and complement 3(C3) in the sera was measured by the ELISA method. Morphology of the pulp and periodontal tissue was investigated pathologically. The results were as follows : 1. In sera, the concentration of IgG decreased gradually upto the 3rd week after the inoculation in the rats of both groups. The concentration began to increase in the 4th week in both groups. In the 5th week, the concentration in the rats immunized was 2.5-fold higher than that in the rats not immunized. 2. In sera, the concentration of IgM in the 1st week was highest, and gradually decreased in both groups. The concentration began to increase slightly in the 5th week in the rats immunized, and in the 4th week in the rats not immunized. 3. In sera, the concentration of C3 in the rats immunized was higher than that in the rats not immunized throughout the experimental period. In the 4th week, the concentration increased slightly in the rats immunized, but decreased in the rats not immunized. 4. The concentration of anti LPS IgM in sera of the rats immunized was high in the 1st week, decreased in the 2nd week, and reached almost plateau values thereafter. 5. The concentration of anti LPS IgG in sera of the rats immunized was high in the 1st week, decreased in the 2nd week, increased slightly in the 3rd week, and decreased again thereafter. In the rats not immunized, the concentration was low for the first two weeks, increased rapidly in the 3rd week and the increase continued for the next two weeks. In the 5th week, the concentration in the rats immunized was similar in value to that in the rats not immunized. 6. In the pathological findings in the rats immunized in the 1st week, intense cellinfiltration in the root pulp, thickening of the periodontal membrane, and slight resorption of the alveolar bone were observed. In the 5th week, necrosis of most of the root pulp and an apical focus, which was surrounded by collagen fibers and connective tissue, were observed. 7. In the pathological findings in the rats not immunized for the first two weeks, cell-infiltration in the root pulp was observed. In the 3rd week, necrosis of about two-thirds of the root pulp, cell-infiltration surrounding the apical crest, and bone resorption of the alverolar bone were observed. In the 5th week, extension of the focus neighboring the apical crest, and intense resorption of the alverolar bone were observed. On the base of these results, the immunization of LPS has the effect of localizing the focus, certainly through the immunological mechanism of anti LPS IgG.
