Rats of wistar strain from the 13th embryonic day to 2 months after birth were used and the neural elements distributed in the tongue were investigated from generation through development chronologically and neurohistologically. The results were as follows : 1. A bundle of nerve fibers entered the tongue on the 13th embryonic day. With growth toward the tongue-tip, each nerve fiber ran more obliquely and its distal end reached the tongue-tip on the 14th embryonic day. 2. On the 13th and 14th embryonic days, most nerve fibers were observed as unmyelinated nerve fibers in a coarse bundle. They gradually became thicker and dense, and more myelinated fibers were found. The nerve fiber bundles accompanying an artery advanced through the lower part of the tongue toward the tongue-tip, branching out, getting smaller in diameter, and meandering. 3. The ganglion cells appeared with a large spherical neucleus and small cytoplasm on the 17th embryonic day. These cells were observed in the muscular layers adjacent to the submandibular ganglion and glands next to the nerve bundle at the tongue-root, and in the base of the vallate papilla. On the 19th embryonic day, these ganglion cells were found scattered in musculars at various sites. Thereafter they increased in number, had large cytoplasm, were either spindle-shaped or star-shaped, and were about matured one month after birth. 4. Lingual ganglia were distributed in glands along the lingual nerve at the tongue-root (submandibular ganglion), in muscular layers near the glands, and in the base of the vallate papilla. Their distribution was relatively fixed. 5. Vallate papilla appeared on the 14th embryonic day. Only one vallate papilla was found located on the posterior lingual median line between the tongue-root and tongue-body. At this stage no nerve fivers were observed under its epithelium. On the 16th embryonic day, a small nerve plexus was found under the epithelium. 6. Fungiform papillae appeared on the 16th embryonic day as epithelial prominence and a number of them were observed at the tongue-body and tongue-tip. On the 19th embryonic day, a small nerve plexus was observed in the papillae. 7. Taste buds appeared in the vallate papilla and fungiform papillae on the first day after birth and reached maturity in four to seven days. 8. Filliform papillae appeared on the 19th embryonic day, starting as proliferation and thickening of the epithelium in the anterior third of the tongue. On the first day after birth, the filliform pappilae had keratinized spindles which curved toward the tongue-root. Connective tissues were found advancing into the mucosal epithelium. 9. On the 14th embryonic day, densely packed and thinly packed cells in the embryonic connective tissue were formed in the midportion of the tongue. On the 17th embryonic day, growth of clusters of young muscular fibers was seen. On the 19th embryonic day, mucosal epithelium, submucosal lamina propria, and muscular layers were differentiated further and distinguishable. Striate patterns on muscle fibers were observed in one to four days after birth. These patterns were matured in one to two months. 10. The posterior lingual glands appeared on the 17th embryonic day as invagination of the epithelial cord into the tongue-root. On the 19th embryonic day, the mucosal cells and serous cells were distinguishable.
