With a view to increasing anti-tumor effect of anti-cancer agents, combination chemotherapy with several anti-cancer agents combined, and combined administration of one or more anti-cancer agents with other medicine have been attempted. Vitamin A, having lysosome labilizer effect and membrane active effect, has been used in combination with one or more anti-cancer agents and increased anti-tumor effect has been reported. While anti-tumor effect is increased by combined administration of Vitamin A, effect on normal tissue is anticipated. However, few studies have been made on this aspect. Therefore, effect of combined administration of Bleomycin (BLM) and Mitomycin C (MMC), most frequently used in oral cancer chemotherapy, with Vitamin A (V・A) on the submandibular gland of domestic rabbits was investigated histopathologically. The results were as follows : 1. In the terminal portion, in comparison with the cases of administration of BLM or MMC only, both cases of BLM-V・A and MMC-V・A combined administration showed marked atrophy and destruction of acinous cells, and pyknosis and deformation of nucleus. Stagnation and hyperemia were observed among scattered acinous cells. Along with these regressive changes, a marked decrease in PAS stainability, PAS positive substance, and A・M stainable substance were observed in B cells (similar to A cells in form and nucleus but contain, in the cytoplasm which stains intensely with eosin, substances which are highly positive to PAS stain and stain red with A・M). These findings were first observed in the peripheral part of the gland and gradually in the middle of the gland as the number of administration increased. In the cases of fifteen dose in particular, these findings were observed markedly. Almost no changes were observed in fine granules of A cells (cylindrical or columnar in cell form and with chromatinrich nucleus and an achromatic net). 2. At the intercalated duct and striated duct, in comparison with the cases of administration of BLM or MMC only, regressive changes such as atrophy of epithelial cells of duct, and pyknosis and deformation of nucleus were observed markedly. Space and rupture of fibrous connective tissue were observed around the ducts. Lumina were markedly narrowed. In the capillaries around the duct, dilation due to stagnation and hyperemia was observed. The foregoing findings were more marked with an increase in the number of administration. 3. At the excretory duct, in comparison with the controls and the cases of administration of BLM or MMC only, atrophy of epithelial cells of duct and pyknosis and deformation nucleus were observed in the fifteen dose cases for both combined administration of BLM-V・A and MMC-V・A. Morphological changes however were slight in the experimental series. 4. Although strong regressive changes resulted presumably from the combined administration of V・A with BLM or MMC, no specific histopathological findings for medicine were found in this experiment. 5. The foregoing findings indicate that BLM and MMC caused regressive changes in the submandibular gland of domestic rabbits and consequent reduction in function. Owing to its pharmacological peculiarity, combined administration of V・A intensified regressive change and promoted reduction in function.
