This study was designed to clarify the possible roles of acid mucopolysaccharides in the wound healing proceess, especially in the formation of dentin-bridge after pulpotomy. Ninety-one pulps in 14 dogs were subjected to pulpotomy and the pulps were dressed with calcium hydroxide. Pulpal healing dynamics were analyzed histochemically. The results may be summarized as follows : 1) The pulp underneath the agent became necrotic soon after the pulpal procedure. This necrotic region showed no toluidine blue metachromasia (pH 4.1 and pH 2.5) and negative staining to alcian blue. 2) The von Kossa's positive layer on the third day after the procedure was positive in metachromasia and also stained by alcian blue. The metachromasia-positive and alcian blue-positive substances were digested by testicular hyaluronidase and chondroitinase ABC but not by streptomyces hyaluronidase. They were assumed to be chondroitin sulfate A and/or C. The richer the fibrous component in this layer, the more scantier they became. 3) Chondrointin sulfate A and/or C as major components, and keratan sulfate as a minor component were detected in the fibrous organic matrix beneath the von Kossa's positive layer since the 7th day after the procedure. This fibrous organic matrix began to be calcified by the 14th day after the procedure, and was assumed to be mainly composed of keratan sulfate. On the other hand, the uncalcified matrix consisted mainly of chondroitin sulfate A and/or C. 4) The acid mucopolysaccharide reactions in the von Kossa's positive and the calcified layer decreased remarkably by the 20th day after the procedure. The above results may suggest that chondroitin sulfate A and/or C and keratan sulfate play an important role in the pulpal calcification.
