FLUORESCENCE LABELING OF PSEUDOURIDINE AND 5-FLUOROURACIL FOR HIGH PERFORMANCE LIQUID CHROMATOGRAPHIC SEPARATION

Reaction with 4-bromomethyl-7-methoxycoumarin(Br-Mmc) allowed the fluorescence labeling of authentic pseudouridine (iJjUrd) and 5-fluorouracil(5FU) using 18-crown-6 as a catalyst. The procedure was rapid, simple and requires little or no experience in derivatization techniques. The reversed phase high performance liquid chromatographic(HPLC) separation of ψ Urd-Mmc and 5FU-Mmc esters was studied with a C_<18> column. Determination limits obtained for ψUrd and 5FU with a new f luorometric detection were 100 pg and 10 pg per 1 Ul injection volume, respectively. Linear calibration plots for peak height versus concentration were up to 10 ng per injection. This fluorescence labeling method for HPLC has an importance in the accurate determination of ψUrd and 5FU which are the most reliable tumor marker and pyrimidine antimetabolite, respectively.
