The percutaneous absorption and the skin metabolism of ^<14>C-ascorbic acid 2-sulfate (AsS) were studied by using male of hartley guinea pigs. A fifty μl of 0.2% AsS aqueous solution was applied occlusively on 5 cm^2 of depilated dorsal skin. When the penetration into the viable epidermis and dermis through the horny layer was expressed as percentage of applied dose, the results obtained were 0.35±0.05% for 2 hours, 1.1±0.1% for 6 hours and 4.8±0.4% for 24 hours after topical application. In vivo skin metabolism of AsS 24 hours after topical application was studied by high performance liquid chromatography. In the horny layer AsS was not converted. The hydrolysis of AsS was found in the viable epidermis and dermis and the conversion rate was 15% for ascorbic acid and 4% for dehydroascorbic acid respectively.
