The immobilization of glucose isomerase was studied. Glucose isomerase from Sterptomyces phaeochromogenus strain SK was entrapped in crosslinked polyacrylamied gel polymerized by γ-ray irradiation or by ammonium persulfate and dimethylamino-propionitrile catalyst.The enzyme entrapped in the gel prepared from 20% acrylamide solution under the irradiation dose 40 krad showed 50-60% activity of the added enzyme. No significant changes by immobilization were observed in optimum teperature and pH of reaction. Heat stability of the entrapped enzyme was considerably fortified and rate of inhibition in the presence of metalic ions such as Cu^<++>, Cd^<++>, and Ni^<++> was somewhat reduced by the immobilization. The entrapped enzyme was not infulenced by the presence of Tris buffer, while native free enzyme was inhibited by the presence of Tris buffer.Continuous isomerization of glucose to fructose was examined using columns containing the immobilized preparations. The conditions were adjusted to obtain a initial isomerization rate greater than 40% and the durability of enzyme activity was investigated. The activity of the column decreased gradually during the course of the operation and diminition probadly depended on the ratio of the amounts of acrylamide and enzyme protein and also on the substrate concentration.
