Cell walls of brewer's yeast were effectively lysed by cell wall lytic enzyme produced by Arthrobacter luteus. Sixty grams of pressed yeast were lysed by 1,600 units of the enzyme within 4.5 hr to give 70% reduction of turbidity. The supernatant obtained from the lysate contained 80% of cell inclusion, and the greater part of the protein in the supernatant had molecular weight of above 50,000. Lacitc acid bacteria grew in medium containing this supernatant and sucrose.Yeast extract was prepared by incubating the above lysate at 45℃ for 11.5 hr. Protease and RNase activities originating from yeast were observed in the lysate during the incubation. This result suggested autolysis of the lysate during the incubation. The yeast extract obtained showed good amino acid balance except for sulfur containing amino acids. The effect of various yeast extract on the growth of lactic acid bacteria was tested. In various samples tested, the yeast extract prepared as above best supported the growth of Lactobacillus arabinosus and Streptococcus faecalis.Yeast extract was prepared from based and sake yeast by the above method except that the yeast cells were lysed by the enzyme in the presence of 2-mercaptoethanol or cysteine.
