In order to confirm the reliability of commercially available Redox Potential electrodes, the following oxidation-reduction systems were adopted to measure the equivalence point potential and Nernst potential respectively : (1) Fe^<++>+Ce^<4+>→Fe^<3+>+Ce^<3+>(2) 6Fe^<++>+Cr_2O_7^=+14H^+→6Fe^<3+>+2Cr^<3+>+7H_2O(3) Quinhydrone electrodel : 2H^++Quinone+2e ⇌ Hydroquinone.Statistical analysis was made based on the series of redox potential measurement involved in the above reactions. The results showed that the reproducibility was fairly good (coefficient of variation <0.91%).The electrode system was then employed to measure the redox potential change in the cultivation broth of yeast (Candida guilliermondii var. formosaensis) using a mixture of n-paraffins as carbon source. Cultivations were carried out bacthwise in a 15 liter jar fermenter whose pH was controlled at 4.0±0.05 automatically. During the course of cultivation, the changes in redox potential and dissolved oxygen tension were recorded simultaneously. Redox potential in this cultivation system responded to the oxygen consumption rate and cell activity. It was also found that in the case of deficiency of substrate, the redox potential increased sharply ; this was utillized in the iterative feedings of the substrate for maintenance of the cell activity and continuation of cell growth.
