Studies on CO_2-fixing Fermentation : (XIV) On the Relationship between L-Malate Formation and Carbohydrate Metabolism of Cell-free Extracts of Schizophyllum commune

Evidences obtained by enzymic studies using crude cell-free extract of Schizo-phyllum commune were as follows.The activity of malic dehydrogenase linked to NAD was strong. The reductive carboxylation of PEP was detected in the presence of Pi, or IDP and KHCO_3.The reductive carboxylation of pyruvate was inhibited by Pi. 3PGA was shown to be more active than pyruvate in the reductive carboxylation system. GAP dehydrogenease activity was detected, and the coupling reaction of malic dehydrogenase with GAP dehydrogenase was found. Aldolase activity was detected, and aldolase could be followed by the reduction of OAA coupled with GAP dehydrogenase.G6P dehydrogenase activity was observed, while it was inhibited by the additions of L-malate and NAD. Both of malic enzyme and lactic dehydrogenase activities could not be found.These evidences indicate that it is affirmative to propose equation (1) and(2)to explaine the process of L-malate fermentation of S. commune.Glucose+2CO_2+4Pi→2_L-malate+2PP…(1)Glucose+2CO_2+2IDP(GDP)+2Pi→2_L-malate+2ITP(GTP)…(2)
