A number of evaluating methods of cellulase preparation were examined. In the past, various procedures to determine cellulase activity have been reported by many investigators, but they stood all on the results obtained by the use of only one substrate. They can not fully evaluate the commericial cellulase products, because so-called cellulase means cellulolytic enzyme system, mixture of many enzymes. In the writer's laboratory, using C.M.C. and filter paper strip as a substrate, reducing power produced on cellulolysis was measured by Somogyi's procedure. And a method which gives the highest reproducibility was established. This method was used in comparing the preparations obtained from the cultures of Trichoderma viride, Aspergillus niger, and Rhizopus javanicus, and differences among them were clearly noticed. At the same time, the procedures measuring physical degradation (break down in shape, decrease in tensile strength, etc.) of filter paper strip were investigated, but none gave reproducible results and were proved unreliable for quantitative determination.
