As the spores of E. ashbyii do not germinate so readily as those of the common molds, the promotive condition of the germination has been studied in order to facilitate the production of riboflavin in a large scale by inoculating the fresh-germinated spores to the fermentation liquor.The following treatment has been found to be most suitable for accelerating the germination of the spores. 35% H_2O_2 was added to a spore suspension in shake culture, adjusting its concentration in the suspension to 1.5〜2%, and so treated spores were immediately inoculated to germination medium. As the germination medium, dilute rice-embryo extract or glucose-peptone medium supplemented with 0.5% corn steep liquor was fitted for practical application.Thus, the riboflavin amount, which was produced in the main fermentation liquor inoculated with 0.02% of the medium containing germinated spores, was not less than that from the culture inoculated with 1% of mycelial suspension and shaken for two days.
