Cotyledon slices of Quercus acutissima CARR. were cultured on agar medium, modified MS (M) or modified N_7(N), supplemented with a combination of 2,4-dichlorophenoxyacetic acid (2.4-D) and 6-benzylaminopurine (BAP), or of 3-indolebutyric acid (IBA) and BAP. After 60 days initial culture, the cultures were transfered to the liquid medium, modified MS with nitrogen at half strength (SC), with/without 0.1 mg/l gibberellin A_3(GA_3), and were shake-cultured. After about 2 weeks, adventitious embryos consisting of cotyledon-, epycotyl-, hypocotyl-, and radicle-like organs were formed in SC medium with 0.1 mg/l GA_3 from M medium with the combination of 1.0 mg/l IBA and 1.0 mg/l BAP, or of 10.0 mg/l IBA and 0.1 mg/l BAP. By culturing on the agar medium, modified HELLER medium, with 1.0 mg/l BAP, these adventitious embryos developed roots and/or shoots, but the shoots had terminal and lateral buds which were translucent and succulent Therefore, nodal segments were isolated from the anomalous shoots, and were cultured by the common method of axillaly bud proliferation. By this culture, normal shoots developed from the nodes. The shoots extended 2 cm or more in length and were transfered to rooting medium and rooted after about 10 days. The plantlets obtained were morphologically complete.
