When cells of Saccharomyces rouxii strain no.K122,a strain used for making miso, were grown on a hypertonic medium and transferred to water, nucleotide-related compounds were released. The nucleotide-related compounds were separated by anion-exchange chromatography and identified by their chromatographic and spectro-photometric behavior and by chemecal analysis.The released meterials consisted largely of 5'-nucleotides, which were considered to have originated in the free pool of the cells. Although isomers other than 5'-nucleotides were present in the intracellular pool, they were not detected in the released materials.From the cells grown on the 2M NaCl-medium, AMP, ADP and ATP were mainly released. On the other hand, from the cells grown on the 2M glucose-medium, UDP-G was released in higher amoung than the other nucleotides. The acid-soluble nucleotide content of cells grown on YM-medium(a hypotonic medium) was less than half that of the hypertonic medium-grown cells.The NaCl-tolerant cells contained considerable amounts of leaky adenyl nucleotides in the intracellular pool.The above results suggest that adenyl nucleotide metabolism may play an important role in the NaCl-tolerance regulation of the yeast.
