The 3rd report Hypertension and Adrenal Cortex The 4th Experiment : The adrenocortical function in experimental salt hypertensive rats.The auther loaded rats with salt to produce hypertension and measured 17-KS excretion in urine compairing with histochemical findings of the adrenal cortex.EXPERIMENTAL PROCEDURES 16 rats, weighing 113g. on the average, were loaded with salt. After 5 months, the rats were divided into three groups, i. e. the first group of increased salt loading, the second group of original experiment, and the third group of withdrawal of salt loading. The experimental period was 192-272 days.RESULT 1) Blood pressure For the first 5 months, an elevation of blood pressure was very slight. The group of increased salt loading showed a marked elevation of blood pressure which after one month reached above 200 mmHg in some rats. In the group of original experiment there was no marked change. In the group of withdrawal the blood pressure was lowerd gradually to the pretreatment level after 40 days.2) 17-KS excretion in urine With the elevation of blood pressure 17-KS excretion in urine was increased. In spite of the maintenance of high pressure it was decreased. At this time when salt loading was increased with resultant hypertension, 17-KS was increased transiently. And when salt loading was withdrawn with resultant lowering blood pressure, it also increased temporarily.3) Histochemical study of the adrenal cortex In rats with long-term salt loading the zona glomerulosa showed a marked atrophy, i. e. hypofunction and the zona fasciculata a hyperfunction. 11-12 days after the withdrawal of salt loading histochemical findings were the same as mentioned above. After 32 days the zona glomerulosa showed a moderate atrophy. After 78-84 days there was observed almost normal finding. In control animals there was no marked change.The 5th Experiment : The adrenocortical functions in experimental pressoreceptor denerved hypertensive dogs.The auther produced experimental pressoreceptor denerved hypertension in dogs, and measured 17-KS excretion in urine compairing with histochemical examination of the adrenal cortex.EXPERIMENTAL PROCEDURE 9 mature dogs weighing 8-16.4kg. were used, in which carotid sinus and aortic nerves on both sides were cut off according to Hering's procedure. They were observed for 3-105 days.RESULTS 1) Blood pressure In all cases the blood pressure was elevated and reached maximal level within 10 days and then decreased gradually with fluctuations.2) 17-KS excretion in urine It was increased immediately after the operation and decreased below the original level. Around the 10th day it, however, returned to the original level. There was no relation to the blood pressure.3) Histochemical study of the adrenal cortex The zona glomerulosa showed hyperplasia, i.e. hyerfunction, but there was no relation to the hypertension.SUMMERY AND CONCLUSION 1) Among 48 patients with various types of hypertension, no hyperfunction of the adrenal cortex was observed except one case of Cushing's syndrome.2) In the experimental renal hypertension, there was no relationship between urinary 17-KS excretion and blood pressure, but hyperplasia of the zona glomerulosa was acknowledged. Therefore, the secretion of hormone of this zona, i. e. sodium-retaining hormone, should be considered to be a very important factor in hypertension.3) In the experimental DCA hypertension, sodium was an absolutely essential factor, which had no relation to urinary 17-KS excretion. The zona glo merulosa showed atrophy.4) In the experimental salt hypertension the blood pressure was elevated parallel to the amount of loaded salt. Urinary 17-KS excretion increased temporarily immediately after the change of salt loading, and had no relation to the blood pressure. The zona glomerulosa showed atrophy.5) In the experimental pressoreceptor denerved hypertension, a transient increase in urinary 17-KS excretion was observed which would be due to stress, but there was no relation to the blood pressure. No hyperfunction of the adrenal cortex was observed, except the zona glomerulosa.
