Purification and Some Properties of a Bacteriolytic Enzyme Produced by Bacillus sp. ML-208

A strain of Bacillus sp. capable of producing a lytic enzyme active toward Micrococcus lysodeikticus and Bacillus megaterium was isolated from soil, and the lytic enzyme was purified about 3,000-fold by salting out with ammonium sulfate, adsorption on CM-cellulose, and affinity chromatography with the lysozyme-lysate of M. lysodeikticus cell-walls. The purified preparation appeared to be homogeneous on disc gel electrophoresis and was most active at pH 6-7,being stable at pH 3.5-9.5. The molecular weight was estimated to be 15,000 dalton by SDS-polyacrylamide gel electrophoresis and the isoelectric point was found to be higher than 10 when checked by isoelectric focusing. Analyses of the enzymatic digestion products from M. lysodeikticus cell-walls indicated that the enzyme was a kind of N-acetylmuramidase.
