Isolation and Characterization of a Gram-positive Methoanol Assimilating Bacterium : Study on the Fermentation of Methanol as a Sole Carbon Source (I)

A facultative methylotrophic gram-positive bacterium was isolated from a soil sample by enrichment cultivation techniques. Based on its taxonomical characteristics it was identified as a new methanol assimilating bacterium and named Corynebacterium methanophilum sp. nov. The bacterium could utilize methanol, thanol, n-propanol, glycerol, acetate, n-propionate, n-butyrate, malate, gluose, fructose, galactose, inositol, sorbitol, mannitol and maltose as sole carbon sources. The bacterium could grow well on methanol at less than 2% (v/v). THe maximum specific growth rate of 0.24 (1/hr) was obtained at 32℃ and pH 7.0 with intermittent feeding of methanol using an automatic feeding device which is controlled by the dissolved oxygen concentration. A biomass of 21.2 (g/l) was obtained after 34hr of cultivation. From 440g methanol and 383.6g oxyge, 208.6g of biomass was produced. The growth yield for methanol was 47.4% in practical operation and was 52% after correction for methanol evaporation during the cultivation. The yield for oxygen was 0.54 (g cell/g O_2). The results of continuous cultivation are also reported. The cells contain 68.4% of crude protein, 10.9% of extractable lipid, 0.6% of crude fiber, 11.7% of carbohydrates and 6.8% of crude ashes. Pepsin difestability of the biomass was 83.3%. Total amino acid content was 56.5% which corresponds to 82.6% of crude protein. The essential amino acid profile is also decsribed.
