Bioconversion of Pimelic Acid into Biotin by Bacillus sphaericus bioB Transformants

Bacillus sphaericus was treated with N-acetylmuramidase and transformed with different bioB expression vectors by protoplast transformations, of which only three could be introduced into B. sphaericus. After cultivating the transformants for 24 h at 28℃, the culture was supplemented with pimelic acid and incubated for 48 h at 37℃. In these conditions, B. sphaericus IFO3525 transformed with a vector containig bioB under the control of the "HpaII promoter" on pUB110 accumulated 20μg of biotin/ml in the medium.
