Large amounts of proteins and UV-absorbing materials were released from salt-tolerant ywast, Zygosaccharomyces rouxii, by osmotic shock. Yeast cells grown in growth medium containing 15% (w/v) NaCl were harvested and suspended in 100 mM NaHCO_3-Na_2CO_3 buffer, pH 9.0,at 30℃ to cause the release of intracellular materials. The amounts of proteins and UV-absorbing materials released by osmotic shock were about one-third of the amount liberated by mechanical disruption or alkaline digestion. Cells subjected to alkaline hypoosmotic shock for 60 min could no longer grow on growth medium with high osmolarity. Compared with untreated cells, the cell volume of shocked cells was smaller and its cell surface was wrinkled.
