A yeast strain, MT1002,isolated from soil, mated and produced spores with Saccharomycopsis lipolytica CBS6124-2 (mating type B) and was identified as S. lipolytica from the taxonomical experimental results.Mutagenesis of MT1002 with N-methyl-N'-nitro-N-nitrosoguanidine (NTG) gave several strains such as SL2-8,MN3-1,etc., all of which exhibited high capabilities of citric acid production.The culture components affecting the citric acid production were investigated with shaking flask cultures. As carbon sources, n-paraffins with C_<14> and C_<15> were sutiable. Limitation of phosphate concentration led to an increase in citric acid production. The presence of cupric ions (0.1-0.2mg/l as CuSO_4・5H_2O) and borate ions (0.1mg/l as Na_2B_4O_7・10H_2O) was found to inhibit the byproduction of isocitric acid and to increase the citric acid production. The miximum productivity of citric acid by muant strain MN_3-1 was 150% (w/w), and the ratio of citric acid to isocitric acid was 89 : 11. Comparisons of enzyme activity among the parent strain and the mutant strains showed that the mutant strains possess higher citrate synthase activity and lower aconitate hydratase activity than the parent, implying that the altered activities may favorably affect the citric acid production of the mutaint strains.
