Immobilization of Nuclease P_1 on Ion Exchange Resins by Titanium Complex Method : Studies on Immobilized Enzymes (II)

Nuclease P_1 was effectively immobilized on ion exchange resins by the titanium complex method. Diaion HPK25,Diaion HPA 10 and Duolite A-4 were suitable as the support. The immobilizing activity depended on the concentration of titanium chloride that was used to prepare the complex with the ion exchange resin. Specific activity of the immobilized nuclease P_1 was 61-62% toward yeast RNA and 71-76% toward 3'-AMP as compared with that of the free enzyme. Such a high expression of specific activity may be related to flexibility of the enzyme remaining after immobilization. A disadvantage of the immobilized enzyme was its relatively low thermal stability, which might be due to the inner environment of the ion exchange resin. The immobilized enzyme was able to perfectly hydrolyze a 1% RNA solution, pH 4.8,over 24 days at 60℃. Moreover, the support was able to be used repeatedly by regeneration.
