Production of Sebacic Acid from n-Decane by Mutants Derived from Torulopsis candida

Strain NC-3-58 was derived from Torulopsis candida 99 as a mutant to produce sebacic acid, and was found to produce sebacic acid to the extent of two and a half times as much as that of the original strani 99. The optimal cultural conditions of sebacic acid production from n-decane by the strain NC-3-58 were investigated both in shaking flasks and in jar fermentors. The rate of sebacic acid production was remarkably affected by the type and content of nitrogen sources, the contents of magnesium sulfate and yeast extract, the temperature of the culture and the rate of aeration. The optimum rate of aeration was found to be relatively low, i.e. 0.16 vvm. Under the optimum conditions thus obtained, 27.3 g/l of sebacic acid was produced from 100 ml/l of n-decane in 83 hr in a 30 l jar fermentor. In the same manner, 33.1 g/l of sebacic acid was produced from 150 ml/l of n-decane in 102 hr of cultivation.
