Effect of Glucose and Ammonium on the Formation of Xanthine Dehydrogenase and Urate Oxidase by Streptomyces cyanogenus : Studies on the Control of Purine Base Metabolism in Streptomyces(III)

The control of formation of xanthine dehydrogenase and urate oxidase by Streptomyces cyanogenus has been studies in cells incubatd in the presence or absence of glucose and ammonium. Changes in concentration of ammonium, cyclic AMP cyclic GMP and in activities of glutamine synthetase and NAD-and NADP-dependent glutamate dehydrogenases of the cells were determined and compared with the formation of xanthine dehydrogenase and urate oxidase. When both glucose and ammonium were added to the medium, uptake and assimilation of ammonium by the cells and accordingly growth of the cells occurred in parallel with repression of the formation of xanthine dehydrogenase and urate oxidase. If glucose or ammonium or both were removed from the medium, the formation of the enzymes occured. In the absence of glucose the uptake and assimilation of ammonium were not observed. The concentration of the cyclic nucleotides and the activities of glutamine synthetase and NAD-and NADp-dependent glutamate dehydrogenases showed no consistent relationship to the regulation of the enzyme formation. The role of the assimilation of external ammonium in the repression of the enzymes is discussed.
