1) Penicillin-protein conjugates prepared in a weakly alkaline saline solution predominantly contained penicilloyl-lysine linkages, and those prepared in a weakly acidic condition mainly contained pheny-lacetylglycyl-lysine linkages. By the method of quantitative precipitin analysis, rabbit antibody combining sites against the former were considered to be differed from those of the latter. 2) By the use of the column chomatography on the specific imunoadsorbent, the antigen combining sites of rabbit anti-BPO-antibodies were shown to be heterogenous. Some antibody populations with different combining sites were revealed to coexist, i.e., combining site against acyl side chain portion of BPO molecule, nucleus portion of BPO molecule and whole BPO molecule containing protein portion surrounding the haptenic group. The values of association constants of these antibody populations with BPO-hpaten were considered to be correlated to the sizes of the cavities of the antigen combining sites. Observations were made on the crossreactivities of rabbit anti-BPO-antibodies with various synthetic penicillins. The crossreactivity of the antibody population against the nucleus portion of BPO molecule with penicilloyl-EACA conjuates of various synthetic penicillins was shown to be complete. However, the strength of crossreactivity of those against the acyl side chain portion of BPO molecule with penicilloyl-EACA conjugates of synthetic penicillins varied with the structural formulas of synthetic penicillins. 3) Patients with hypersensitivity to penicillin were skin-tested with BPO-hapten and its related compounds. By statistical analyses of their wheal responses, the antigen combining sites of human skin senistizing antibodies to penicillin were considered to be heterogenous.
