The role of FSH on steroidogenesis in immature rat ovaries was investigated with reference to cyclic AMP and protein kinase in vitro. The ovarian cyclic AMP level was increased by both FSH and LH, and the effect was much potentiated in the presence of aminophylline. Protein kinase activity was also stimulated by FSH. The amount of LH contained in the FSH preparation used appeared to be insufficient to account for the stimulating activity. In regard to steroidogenesis, estradiol-17β and progesterone produced by the ovaries during incubation with gonadotropins were measured. Estradiol-17β production by the ovaries was markedly increased mainly by FSH, whereas progesterone production was more stimulated by LH. Furthermore, addition of dibutyryl cyclic AMP in the medium resulted in a significant increase in both estradiol-17β and progesterone. The present results suggest that FSH as well as LH stimulate steroid biosynthesis by activating adenyl cyclase, cyclic AMP and cyclic AMP-dependent protein kinase.
